Research interests
Claudio Villani is full professor of Organic Chemistry at University of Roma La Sapienza , Chemistry and Technology of Drugs Department. His main interests are in the field of organic stereochemistry, with emphasis on the application of chromatography and spectroscopy (NMR, CD, MS) to problems in static and dynamic stereochemistry. He has been involved in the design, synthesis, characterization and commercialization of chiral stationary phases for HPLC applications.

Scientific achievements
Claudio Villani obtained his Laurea degree in Pharmaceutical Chemistry and Technology in 1984 in the group of Francesco Gasparrini, working on the synthesis of silica based chiral stationary phases (CSPs) for HPLC applications. In the following years, he continued to work on the same topic, developing a series of CSPs incorporating small synthetic chiral molecules (Chromatographia 1987, JCA 1988), synthetic macrocycles (JOC 1995, 1997), macrocyclic antibiotics (Anal Chem 2000, JCA 1999), polyamides (JCA 2005). Part of the activity was done at the School of Chemical Science-University of Urbana Champaign working with prof. W.H. Pirkle during the years 1993-94. Chiral-racemic or non-chiral HPLC silica based hybrid materials incorporating fullerenes (JACS 1997) or carbon nanotubes (Carbon 2006) were prepared and used to study the interaction of the carbon fragments with several guest probes. Recent activity in the field has seen the first introduction of small particles for ultra-high performance applications (JCA 2010, Anal chem 2012, TrAC 2014). Insights of the molecular recognition mechanisms that govern enantioselectivity in LC systems have been derived from free solution studies and molecular modelling (J. Comp. Chem. 2000, 2007, EurJOC 2011). Parallel studies in the gas phase were subsequently performed using small macrocyclic amide receptors that showed unprecedented enantioslectivity towards small aminoacidic guests. Investigations in this area were conducted on electrospray generated proton bound host-guest dimers and trimers and observing their gas phase behavior upon collision induced dissociation or gas-phase guest exchange reactions (JACS 2005, 2008; Chem Commun 2009, Chem Eur J 2011). Claudio Villani gave a substantial contribution to the stereodynamic investigation of chiral species, including drugs or drug like molecules (JACS 1992, 2000; JOC 1995; Tet Asym 1997, Chem Commun 2012; JCA 2014), featuring labile stereogenic elements. A combination of techniques are used to extract kinetic data, including on-column dynamic HPLC, dynamic NMR, off-column thermal isomerization, stopped-flow CD. These studies have direct relevance both in setting and consolidating experimental approaches, and in providing dynamic structural details of chiral species.
