This module of Elements of Physical Chemistry aims at providing the Biotechnology student with basic knowledge of spectroscopy and the fundamental theoretical tools to understand and correctly interpret the application of some physical-chemical instrumental techniques in the analysis and the structural characterization of systems of biotechnological interest. At the end of the course the student should also be also to solve simple numerical problems and to apply the acquired knowledge in the subsequent courses.

 

 

Prerequisites:

Basics knowledge of chemistry (atomic model, chemical bond, molecular structure) and of mathematics for the solution of simple equations. Basic knowledge of biochemistry can be useful to better understand some examples of application but do not represent a prerequisite.

 


Topics:

Introduction to spectroscopy: the electromagnetic radiation, the quantization of energy, the regions of the elctromagnetic spectrum, the intensity of spectra.

Basics of molecular rotational states.

Molecular vibrations and infrared spectroscopy.

Electronic states and UV-visible spectroscopy.

Nuclear magnetic resonance and introduction to its application for molecular identification.

Elements of fluorescence, circular dichroism and Raman scattering.


 

Examination:

The exam will be a written test (minimum points to pass 18) lasting maximum two hours in which there will be four excercises to be solved and commented about the basics of spectroscopy and on the fundamental spectroscopy techniques studied in the course (UV-visible absorption, NMR, IR).


 

The course will be carried out as lectures in which the professor will also take advantage of slides (that will be available on e-learning) and of the blackboard to illustrate the contents, mentioning the bibliography used as reference that can be used as material for further in-depth study. Some of the lectures (about 15%) will be dedicated to solving excercises and numerical problems to strenghten the knowledge, to show applicaton examples and to facilitate the preparation for the final examination.

 

Attendance is highly recommended but not mandatory. The sessions dedicated to the solution of problems and numerical excercises will be particularly useful for the final examination.

Alessandra Del Giudice è nata nel 1988 a San Severo (FG). Ha conseguito con lode la laurea triennale in chimica nel 2011 e la laurea magistrale in chimica, indirizzo inorganico-chimico fisico, nel 2013 presso l'Università di Roma La Sapienza. Nel 2013 è stata abilitata all'esercizio della professione di chimico. Nel dicembre 2016 ha conseguito il titolo di dottore di ricerca in Scienze Chimiche nella stessa università, dove ha lavorato come assegnista di ricerca dal 2017 e, dal 2022, come RTD-A. Il suo lavoro durante il dottorato si è incentrato sull'applicazione di tecniche di scattering in soluzione e spettroscopie ottiche per studiare processi che coinvolgono le proteine, come la denaturazione e l'oligomerizzazione. Ciò ha previsto anche dei periodi di visita presso l'Università di Lund (Svezia). I suoi interessi di ricerca attuali includono anche la caratterizzazione strutturale di sistemi "soft-matter", in particolare formati dall'autoassociazione di molecole anfifile. Durante il suo lavoro di ricerca ha avuto la possibilità di partecipare a numerosi esperimenti di "small angle X-ray scattering" con radiazione di sincrotrone. In aggiunta, Alessandra collabora alla gestione del SAXSlab Sapienza fornendo assistenza tecnica e supporto scientifico agli utenti per realizzare misure di small/wide angle X-ray scattering in laboratorio.
