Part XI – Research Activities
Research statement
My main interest lies in understanding how neuronal networks process information to enable proper brain function. During my PhD, I became deeply fascinated by the extraordinary role exerted by glial cells in supporting and shaping the correct development and maturation of synapses. A role that remains crucial for maintaining network functionality throughout life. I gained substantial expertise in electrophysiology and time-lapse imaging, which allowed me to investigate how microglia influence hippocampal synaptic transmission under both physiological and pathological conditions. In recent years, I have been focusing on the study of neurodevelopmental disorders, investigating the contribution of different high-risk genes to the pathophysiology of autism spectrum disorders (ASD). I have investigated how mutations in high-risk ASD genes affect neuronal network plasticity, aiming to identify convergent molecular pathways and explore the reversibility of ASD-like phenotypes. My postdoctoral training has expanded my expertise, ranging from electrophysiology, confocal microscopy, behaviour, molecular biology and sequencing. Currently, I am studying SYNGAP1, a gene critical for synaptic plasticity and cognitive function, whose mutations are strongly linked to intellectual disabilities and ASD. By integrating genetic approaches with human-derived in vitro models, I seek to elucidate the impaired molecular mechanisms driving these conditions and identify novel targets for therapeutic intervention. Looking ahead, I am committed to further dissecting the dynamic processes of neuroplasticity, from early nervous system development to the formation of functional neuronal networks, bridging molecular insights with translational applications to advance treatments for neurodevelopmental disorders.
Organization, direction and coordination of research groups
Research Topic: Translational study on SYNGAP1 mutations associated with developmental and epileptic encephalopathy and autism spectrum disorder.
I am the scientific lead and coordinator of an ongoing translational research initiative focused on the SYNGAP1-related disorder, a rare genetic disease characterized by developmental and epileptic encephalopathy (DEE) and autism spectrum disorder (ASD). My responsibilities include overseeing the research activities and ensuring effective coordination among families, clinicians, and research teams. This project is rooted in a multidisciplinary and collaborative framework that bridges basic science, clinical care, and patient advocacy. Institutions involved and under coordination: Università Sapienza di Roma; Centro di Ricerca in Neurobiologia “Daniel Bovet”; Istituto Italiano di Tecnologia (Prof. Giancarlo Ruocco, Dr. Mattia Miotto); Policlinico Umberto I – Pediatria (Prof. Alberto Spalice); Fondazione IRCCS Casa Sollievo della Sofferenza (Prof. Jessica Rosati); Fondazione IRCCS Istituto Neurologico Carlo Besta (Prof. Stefano D’Arrigo); APS Famiglie SYNGAP1 Italia; APS Le Note di Fra. This activity is also supported by private donations, for which I am the named recipient, from the patient advocacy groups APS Famiglie SYNGAP1 Italia and APS Le Note di Fra, recognizing the scientific and organizational value of the project.
Research Topic: Role of microglia in synaptic development and function.
I lead and coordinate research activities focused on neuron–microglia crosstalk and its role in synaptic maturation within the hippocampus. My work is contributing to define the impact of Cx3cl1/Cx3cr1 signaling on microglial physiology and neurotransmission. These projects involve coordination and collaborative efforts with multiple research groups, including those led by Prof. Cristina Limatola (Dept. Physiology and Pharmacology, Sapienza University of Rome), Dr. Cornelius Gross (EMBL Rome), and Prof. Marie-Eve Tremblay (University of Victoria, Canada). In this context, I co-authored over 10 publications on neuron–microglia interactions across different in vivo models.
Research Topic: Genetic basis of neurodevelopmental disorders (NDDs).
As a postdoctoral researcher in the laboratory of Prof. Gaia Novarino (IST Austria), I led and contributed to several studies aimed at uncovering the molecular mechanisms underlying neurodevelopmental disorders. These studies required careful experimental design and collaborative efforts across multiple research centers and disciplines. Institutions involved and under coordination: Institute of Science and Technology Austria; Medical University of Vienna (Prof. Igor Adameyko); Center for Molecular Medicine (Dr. Christoph Bock).
Research Topic: Rewiring of the brain via the gut-brain axis.
I lead and coordinate ongoing research activities investigating how peripheral inflammation, particularly of intestinal origin, impacts neuronal network function and plasticity. My role involves supervising experimental design and implementation, as well as ensuring effective collaboration and communication among the participating research teams. This multidisciplinary project brings together expertise from different institutions, and I coordinate the efforts of the following research groups: Prof. Fabio Sciubba (Dept. of Environmental Biology, Sapienza University of Rome); Prof. Giuseppe Esposito (Dept. of Physiology and Pharmacology, Sapienza University of Rome); Dr. Fabrizio Antonangeli (National Research Council, Italy); Università degli Studi LINK (Prof. Maria Rosito). The overarching goal is to elucidate the molecular and cellular mechanisms linking gut-derived inflammatory signals to brain function, with implications for neurodevelopmental and neuropsychiatric disorders.
