Technical skills and competences
Electron microscopy techniques. Methods used and created
1. Combined use of saponin, ruthenium red, osmium, thiocarbohydrazide, (Sap-RR-Os- Th method) to
highlight the fine structure of the extracellular matrices, in ovarian follicle and in the oocyte, with the
scanning and transmission electron microscopy. This method has been applied by other groups that,
in the world, work on this field.
2. Sound and ultrasound adjustable system, able to softly clean the samples surfaces for the
observation at the scanning electron microscope.
3. In addiction to those above mentioned, prof. Familiari usually uses in his routine laboratory work the
following techniques: immunofluorescence, ruthenium red and Lanthanium in transmission electron
microscopy, scanning electron microscopy, laser confocal microscopy.
